چکیده: Abstract: Using electrospinning methods in the field of manufacturing scaffolds used in the field of bone tissue engineering due to porous structure and fiber-like structure, has remarkably been noticed. In this study, polycaprolactane polymer (PCL) was used as a matrix and a biodegradable polymer with good tensile and mechanical properties. The release and presence of bovine serum albumin (BSA) protein distributed in the PCL matrix through electrospinning blending method were also investigated. The presence of hydroxyapatite (HA) particles was placed inside the PCL matrix to aid in better ossification, release and better biocompatibility of the scaffolds according to the protein. Their microstructure, molecular groups, wettability, swelling, and mechanical properties were also determined. The results of SEM and FTIR characterization experiments showed that HA and BSA proteins can be included in PCL fibers without significant effect on the main molecular groups of PCL structure. The combination of both HA and BSA protein can reduce the hydrophobicity of PCL scaffold by about 13 ° for bone regeneration. Also in the study of swelling and degradation of scaffold fibers for 100 days in PBS solution, showed that the swelling of scaffolds containing the above components has an increasing trend. After about 40 days, the PCL scaffold began to degrade and its swelling decreased. The mechanical properties of scaffolds were investigated in dry and wet conditions (using PBS solution). BSA release kinetics from hybrid scaffolds were investigated. Higuchi kinetic model was the most appropriate model to explain protein release. SBF immersion test was generally performed to evaluate the formation of apatite on scaffolds. Changes in the atomic ratio of Ca / P during the immersion period were studied for 7 days. The results showed that due to the synergistic effect of HA and BSA protein, better ossification was performed on the hybrid scaffold PCL-HA-BSA. Scaffold compatibility was evaluated by MG-63 osteosarcoma cells and showed the remarkable ability of PCL-HA-BSA hybrid scaffold to regenerate bone. Interactions between BSA and HA proteins at 170 ns were investigated using GROMACS molecular dynamics simulation software. The results showed that the protein molecule reacted physically from its hydrophilic sites with the help of hydrogen bonds with HA ions; Which indicates a tendency to bring these two components closer
